The effect of zinc oxide nanoparticles on the prooxidant and proapoptic effects of glyphosate

Анотація

The specific feature of nanoparticles to increase the transport of chemicals into cells through the membranes makes the problem of potentiating the toxic effect of chemical contaminants when they enter the tissues and organs of the body together with nanoparticles an urgent issue. The purpose of this study was to evaluate the integral effect of zinc oxide nanoparticles and the chemical toxicant glyphosate on the formation of oxygen active forms and the processes of apoptosis and necrosis of blood neutrophils. Research methods. Male rats were administered intragastrically with 0.5 ml of a ZnO nanoparticles suspension at a dose of 100 mg/kg and glyphosate (in the form of roundup herbicide) at a dose of 250 mg/kg of body weight for 14 days. Toxicants were administered simultaneously and separately. On the 15th day the formation of reactive oxygen species, the number of cells with a reduced mitochondrial transmembrane potential, and the intensity of apoptosis and necrosis were measured in blood neutrophils. Results and discussion. It was established that the maximum changes of the intensity of release of oxygen active forms, the number of cells with a reduced mitochondrial transmembrane potential, and the level of blood neutrophils with apoptotic and necrotic changes occured in the group of animals that were injected with ZnO nanoparticles together with the glyphosate. It should be noted that in animals of this group all indicators increased significantly compared to rats that were administered with only herbicide. Conclusion. Zinc oxide nanoparticles enhance the ability of the herbicide glyphosate to enhance the generation of intracellular active forms of oxygen, increase the number of cells with a reduced mitochondrial transmembrane potential, as well as the level of blood neutrophils with apoptic and necrotic changes.
