Експериментальна та клінічна фізіологія і біохіміяThe article presents the results of the study of transforming growth factor β1 (TGF-β1) production in rats with chronic toxic hepatitis. TGF-β1 is the main profibrotic cytokine produced by Kupffer’s cells, stellate cells, sinusoidal endotheliocytes and secreted by platelets and hepatocytes. TGF-β1 stimulates expression of matrix genes, and reduces production of matrix metalloprotease and increases production of tissue inhibitor and initiates apoptosis of hepatocytes and inhibits proliferation of liver cells. Activation of fibrogenes is usually accompanied by the decreasing of signs of inflammation and leads to disruption of the connective tissue structure of the liver, which then determines the functional failure processes of regeneration of liver parenchyma.
The aim of the study was to find out the features of TGF-β1 production in rats with chronic toxic hepatitis as a factor that affects the processes of reparative regeneration of liver tissue.
The research was conducted on 96 laboratory rats. The main group of the study consists from 48 rats, which according to the age were distributed to three subgroups. First subgroup included 16 young immature rats aged 2 months, second subgroup – 16 young mature rats aged 6 months. Third subgroup was represented by 16 old mature rats aged 18–20 months. The control group included 48 intact rats of the same age, which was also divided into three subgroups.
On the rats of the main group was experimentally reproduced model of chronic toxic hepatitis. To modulate chronic toxic hepatitis for ten weeks intragastric 20 % CCl4 oil solution in dose 0,1 ml/100 g was administered three times a week in combination with a 5 % ethanol solution (orally).
After the experiment the euthanasia of animals was carried out under ketamine anesthesia (0,1 mg/100 g weight, intramuscularly) by decapitation. Content of transforming growth factor β1 was determined by enzyme-linked immunosorbent assay using highly sensitive sets of the firm DRG (Germany).
Statistical analysis of the obtained data was conducted with using of the program Statistica 6,1 (license number AXXR910A374605FA) by nonparametric methods of the study according to redistribution of the data (criterion Mann – Whitney).
After evaluation of the difference in content of TGF-1β between main and control groups the following data was observed: the group of young immature rats with chronic toxic hepatitis has a statistically significant increasing of TGF-1β content by 6,5 % compared with the group of intact animals of similar age (147,9 ± 7,56 vs 138,8 ± 13,2 pg/ml, p < 0,05). In the analysis of this indicator in the group of young mature rats with chronic toxic hepatitis observed its growth to 1,07 times compared with animals from the control group of the same age (148,9 ± 11,05 to 139,0 ± 7,3 pg/ ml, p < 0,05). In the group of older mature rats with chronic toxic hepatitis content of transforming growth factor β was at 7,85 % more than in the same age group of intact animals (155,3 ± 9,54 to 144,04 ± 7,38 pg/ml, p < 0,05).
After the analyses of the data ther following conclusions can be made: the concentration of transforming growth factor 1β does not dependent on the age and the increasing of its production can be observed in the case of prolonged action of pathogenic factors.
