Experimental evaluation of the impact of L-lysine aescinat the reproductive function of rats

Анотація

The pilot research of substance of a L-lysine aescinat for an assessment of gonadotoxic and embriotoxic action is conducted. The condition of gonads was studied on white nonlinear male rats. Macroscopic indicators of seminal glands, namely visual indentification of pathology, measurement of their weight and the size were conrolled. The assessment of a functional condition of spermatozoa was carried out on the following indicators: concentration of spermatozoa in Goryaev,s camera, calculation of quantity of pathological forms and dead spermatozoa, peak of their mobility, osmotic and acid resistance. The condition of sperm fertile epithelium of rats was estimated by quantity of structurally functional elements (total quantity of normal spermatogonia, quantity of tubules with desquamated epithelium seed forming). Embryotoxic effect of a preparation was studied on female rats. Calculation of yellow bodies, places of implantation, live embryos, places of resorption, general embryonic mortality, before- and after-implantation mortality, fetal weight, the cranio-caudal size, weight and diameter of placenta was carried out.

Based on the results of the experiment gonadotoxic action revealed that L-lysine aescinat doses 15,4 mg/kg (1/50 DL50) and 3,1 mg/kg (1/250 DL50) does not change the size and weight coefficient does not cause significant changes in functional parameters of spermatogenesis in rats, but has effect on the process. Sperm count in rats of the first and second experimental groups compared with the control group was reduced by 4,40% and 2,25%, with an increase in abnormal sperm forms and the number of dead sperm.

Effect of L-lysine aescinat results in increased sperm motility compared with control respectively 35,0% and 21,0%, reduced osmotic respectively 6,60% and 4,65%, and acid respectively 6,56% and 2,70%, consequently the resistance of spermatozoa.

There was a decrease of redox processes of sperm in the first experimental group was 39,33%, and the second experimental group – 23,68% compared with control.

The state of sperm fertile epithelium of rats from the research groups was characterized by a decrease in the number of normal spermatogonia respectively 1,67% and 0,55%. Index of spermatogenesis in the first experimental group was (3,40±0,82 SU) in the second experimental group (3,54±1,10 SU), that was less than the control group (3,75±0,69SU) by 9,33% and 5,60%.

The daily observation of pregnant rat females did not show their death during the experiment in either group. The introduction of the drug L-lysine aescinat did not effect on the duration of the pregnancy female rats, but there were records of abortion and premature birth. In groups where the animals received the drug at doses of 1/50 DL50 (15,4 mg/kg) and 1/250 DL50 (3,1 mg/kg) in pregnant females there was no clinical picture of poisoning. The body weight of animals has increased during the experiment. In the course of the macroscopic review of female fetuses that received L-lysine aescinat during the entire pregnancy, it was found that in all embrios of the control and experimental groups no facial or cranial malformations were found. Cranium itself was of oval-oblong shape. Pinna and eyelids were closed. The anterior abdominal wall was closed, with no signs of umbilical hernia. Tail length was usual. Well-developed limbs, without pathologies.

Indicators of before and after-implantation mortality and general embryonic mortality in the general experimental groups had probable differences from that of the control.

In experimental animals fetal weight and cranio-caudal size, weight and diameter of placenta had no significant difference from that of control animals. The external malformations were not seen.

It has been established that a L-lysine aescinat does not influence generative function of rats.
