Influence of liposomal form of quercetin on lipid metabolism and indicators of the liver state in experimental diabetes mellitus type 2

Type II diabetes is one of the most important problems of modern medicine. The development of type II diabetes is accompanied by prolonged disturbances of carbohydrate, lipid and protein metabolism, leading to pathological changes in the functioning of various organs and systems. Hyperglycemia plays a leading role in the mechanisms of chronic complications of diabetes. In this regard, for compensation of metabolic processes in diabetes it is important to normalize not only the level of glucose but also lipid metabolism. Our researches were performed on non-linear white male rats (body weight 200–250 g) which were divided into three groups: 1 – control (intact animals), 2 – animals with diabetes type II, III – at the basis of diabetes animals were injected with liposomal form of quercetin (Lipoflavon ) (10mg / kg, intraperitoneally, daily). Experimental treatment started in 10 weeks after the administration of streptozotocin (STZ), for 2 weeks. Single injection of streptozotocine (STZ, Sigma, USA, 30 mg/kg body weight) was administered intraperitoneally to experimental animals after the previous 4-week high fat diet. STZ was dissolved immediately before the administration in 0,1 molar citrate buffer (pH 4,5); control group was injected with the appropriate amount of citrate buffer. Further experiments included only animals with glucose levels no lower than 10.8 mmol/l in 2 weeks after STZ administration. The control group of rats was kept on a standard diet. All the principles of the European Convention for the protection of laboratory animals were used.

It was shown that diabetes type II accompanied with marked changes in carbohydrate and lipid metabolism. In particular increase of glucose and glycated hemoglobin levels in blood was observed, which may be the result of a decrease in glucose transport into cells of muscle and adipose tissue, impaired intracellular glucose metabolism, reduction of waste, reduced synthesis and increased breakdown of glycogen in the liver. There was increased activity of alanine and aspartate aminotransferase, alkaline phosphatase. In studying the parameters of lipid metabolism in animals with type II diabetes the dyslipoproteinemia was registered: increased concentrations of total cholesterol, triglycerides, low-density lipoproteins, reducing the concentration of high density lipoproteins in the blood serum. The established biochemical changes in blood parameters in experimental diabetes type II indicate significant dysfunction of various organs and systems in general agreement with the clinical course of the disease and may be an additional confirmation of the formation of stable type II diabetes in experimental animals.

The results give reason to believe that the liposomal form of quercetin reduces glucose levels and glycated hemoglobin, decreases the activity of aminotransferases and alkaline phosphatase, the level of bilirubin, total cholesterol, triglycerides, low-density lipoprotein levels in blood serum with concomitant increase of HDL level. The preparation of quercetin Lipoflavon positive influence on carbohydrate and lipid metabolism is due to its antioxidant, antiradical, anti-inflammatory and detoxifying effects.
